It was founded on December 20, 1798, from parts of Warren and Green Counties. It was named for the Barrens, the meadowlands that cover the northern third. As of 2010 the population was 42,173. Its county seat is Glasgow, Kentucky. The county is part of the Glasgow Micropolitan Statistical Area.

In 2007, Barren County was named the number one rural place to live by Progressive Farmer magazine. Glasgow is the principal city of the Glasgow Micropolitan Statistical Area, which includes all of Barren and Metcalfe counties.

WWKU is a radio station licensed to Plum Springs, Kentucky, USA. The station is currently owned by Newberry Broadcasting, Inc. and runs ESPN Radio sports programming. The station moved from 1440 to 1450 in May 2008 to allow for an upgrade to a stronger signal of 1,000 watts day and night (compared with the 500 watts day and 30 watts night it previously had). Before having Plum Springs as its official city of license, WWKU was licensed by the FCC to the town of Glasgow, Kentucky.
